Ok, everyone on this board knows that I love european cars but you're about to make a HUGE mistake by buying ANY of those cars you've listed.
If you think that any of them are going to be anywhere close to as reliable as the Ford Flex you already own I've got a bridge to sell you. I also take paypal. Just saying. I want to make it easy for you. Looking out, ya know. Friends and family payment only.
All of them will drain your wallet and in a hurry. The Benz and the Audi faster than the Volvo but it's not like the Volvo will be far behind.
Get over whatever hang up's you have and go buy yourself a minivan or a Tahoe or a Suburban or a Sequoia.
The r500 has the m113 engine. Used in just about anything v8 Mercedes for a long time. They're reliable and so are the trans. Parts aren't expensive and easy to find. I'm not sure how easy it is to access in the r-class but I'd drive an m113 powered Benz with the same piece of mind as a ls or Lexus v8 powered vehicle. But the r350, I'd avoid lol.
Myself and some folks here think I'm a Benz fanatic so take my advice with a grain of salt. I'm right though.

Also, the answer is suburban. You may not like it, but it'll do 125% of everything you need, and run twice as long as any of your listed options, and have 1/4 the repair costs. Plus, no need to LS swap. Want speed? Boost. Want to tow something? Anything? Ever? Once? Want to go off road on a whim? 4 kids? Easy. 6 kids. Easy. Engine booms? Every jy and half the guys on your street have a replacement.
Get a dog. Get two. It is a do anything and everything vehicle, and cheap as anything. And runs great on 87.

Having ridden in my sister's Suburban many times - she's a Burban fanatic- there is no way a Suburban is hauling 6 kids unless the driver is alone and all 6 kids are just old enough to be out of booster seats but before the earliest onset of puberty.

I think he wants 6 total. Not 6 kids. My sister has a recent one and hauled a family of 6 quite often. I can't speak to their comfort level because I haven't been in on the full loads.
Of his choices the Mercedes might be the best at hauling 6 because it's the most minivan like.
How about this? A 740 Turbo would be fun and they are pretty reliable. :)

This is where the aftermarket can help families with a pickup truck and more than three kids. Mount two seats and seatbelts facing backward in the bed of a pickup truck. Brat style! Hell, adapt a fifth wheel hitch mounting setup and make 'em removable.
And
All 6 of us had plenty of room and comfort, made it non stop, and took everything we wanted.
So the answer was the Flex I already owned.
I thought you were seeing awful fuel economy? What changed? Numbers look great and no leak looks even better.
It's almost always cheaper to fix what you own, especially with those alternatives.
Glad to see the flex is back.

Well I changed the oil and plugs, did an intake valve cleaning service, and ran some fuel cleaner through it.

My 01 Burb took 2 adults and 6 kids (ages 14, 13, 13, 11, 9 and 7) on a two weeks vacation in Florida in this past January. That was a 3200 miles round trip for us.
While we did need to bring the Thule cargo carrier for the lighter luggage, everyone was very comfortable for the entire trip. It tackled NYC twice on that trip, Washington and the Florida heat.
Aaaand made it home perfectly fine with no issues. Still hasn't leaked a drop.
Now let's get back into it.
V8. Manual. Wagon. A holy combination if there ever was one- which car objectively would be the best? I've heard many sing the praises of the Mercedes V8s, but the Volvo V8s were Yamaha-assisted mills like my 4A.
