I'm going today to pick up a craigslist mega-squirt with an unknown background. I don't have a simulator. Is there a way to test it with a multi meter, or at least show that it powers on?

You could hook up power to it and try to connect to it with a computer, if that works then at least you'd know it's not dead as a doorpost.
Power and ground and computer would be easiest way.
You could also take a multimeter and check to see if the 5V refererences have power.
