Inside, unheated on a concrete floor. I can't put it on jackstands in case the worst happens and it has to be moved quickly.

To protect the tires, would you put somethig between the floor and the tires?

Thick plastic?

I would just make a point to rotate/move the tires every once in a while, i.e. jack up one side and turn them. The original tires on my trailer sat quite a bit and they didn't have any flat spots. They are radials not bias ply so that may account for some of it.

I use scraps of OSB, which is all it's really good for.

I don't see any advantage to putting something between the tires and the concrete. I was under the impression that you want to take the load from the tires to avoid flat-spotting them or putting a "set" in the radial plies. Other than than, the tread surface should react the same to wood, plastic or concrete.

How about two of these:

Back up into them and they raise the trailer off the ground, but you can pull forward quickly and be on your way. (Zombie attack?)

Put it on old cinder blocks. If you have to go in a hurry hit the blocks with a sledge, shatter them and haul ass.
